RESUMO

We present a highly deuterium atom economical approach for the synthesis of deuterated alcohols via the single electron transfer (SET) reductive deuteration of acyl chlorides. Cost-effective sodium dispersion and EtOD-d1 were used as the single electron donor and deuterium donor, respectively. Our approach achieved up to 49% deuterium atom economy, which represents the highest deuterium atom economy yet achieved in SET reductive deuteration reactions. With all 20 tested substrates, excellent regioselectivity and >92% deuterium incorporations were obtained. Furthermore, we demonstrated the potential of this methodology by synthesizing four deuterated analogues of pesticides.

RESUMO

Through sustainable development goals 3 and 8 and other policies, countries have committed to protect and promote workers' health by reducing the work-related burden of disease. To monitor progress on these commitments, indicators that capture the work-related burden of disease should be available for monitoring workers' health and sustainable development. The World Health Organization and the International Labour Organization estimate that only 363 283 (19%) of 1 879 890 work-related deaths globally in 2016 were due to injuries, whereas 1 516 607 (81%) deaths were due to diseases. Most monitoring systems focusing on workers' health or sustainable development, such as the global indicator framework for the sustainable development goals, include an indicator on the burden of occupational injuries. Few such systems, however, have an indicator on the burden of work-related diseases. To address this gap, we present a new global indicator: mortality rate from diseases attributable to selected occupational risk factors, by disease, risk factor, sex and age group. We outline the policy rationale of the indicator, describe its data sources and methods of calculation, and report and analyse the official indicator for 183 countries. We also provide examples of the use of the indicator in national workers' health monitoring systems and highlight the indicator's strengths and limitations. We conclude that integrating the new indicator into monitoring systems will provide more comprehensive and accurate surveillance of workers' health, and allow harmonization across global, regional and national monitoring systems. Inequalities in workers' health can be analysed and the evidence base can be improved towards more effective policy and systems on workers' health.

It is challenging to conduct groundwater contamination risk assessment in fractured aquifers containing a large number of complex fractures, especially in a situation where the uncertainty of massive fractures and fluid-rock interactions is inevitable. In this study, a novel probabilistic assessment framework based on discrete fracture network (DFN) modeling is proposed to assess the uncertainty of groundwater contamination in fractured aquifers. The Monte Carlo simulation technique is employed to quantify the uncertainty of fracture geometry, and the environmental and health risks of the contaminated site are probabilistically analyzed in conjunction with the water quality index (WQI) and hazard index (HI). The results show that the contaminant transport behavior in fractured aquifers can be strongly affected by the distribution of the fracture network. The proposed framework of groundwater contamination risk assessment is capable of practically accounting for the uncertainties involved in the mass transport process and effectively assessing the contamination risk of fractured aquifers.

RESUMO

Microplastics (MPs) exposure generally triggers oxidative stress in fish species and vertebrate pigmentation is commonly influenced by oxidative stress, but MPs-induced oxidative stress on fish pigmentation and body color phenotype has not been reported. The aim of this study is to determine whether astaxanthin could mitigate the oxidative stress caused by MPs but at the expense of reduced skin pigmentation in fish. Here, we induced oxidative stress in discus fish (red skin color) by 40 or 400 items/L MPs under both astaxanthin (ASX) deprivation and supplementation. We found that lightness (L*) and redness (a*) values of fish skin were significantly inhibited by MPs under ASX deprivation. Moreover, MPs exposure significantly reduced ASX deposition in fish skin. The total antioxidant capacity (T-AOC) and superoxide dismutase (SOD) activity in fish liver and skin were both significantly increased with the increase of MPs concentration, but content of glutathione (GSH) in fish skin showed a significant decrease. For ASX supplementation, the L*, a* values and ASX deposition were significantly improved by ASX, including the skin of MPs-exposed fish. The T-AOC and SOD levels changed non-significantly in fish liver and skin under the interaction of MPs and ASX, but ASX significantly reduced GSH content in fish liver. Biomarker response index indicated that ASX could improve the moderately altered antioxidant defense status of MPs-exposed fish. This study suggests that the oxidative stress caused by MPs was mitigated by ASX but at expense of reduced fish skin pigmentation.

RESUMO

OBJECTIVE: To evaluate the risk of noise induced hearing loss among workers in petrochemical enterprises. METHODS: Number of workers exposed to noise were recruited from three petrochemical enterprises. The noise exposure level(L_(EX, W)) of the research objects was measured, their occupational history was investigated, and the audiometric testing was carried out. The ISO 1999:2013 model was used to calculate the change of hearing threshold level and the risk of hearing loss in each post, and compared the result of model with the result of the audiometric testing. RESULTS: The median of L_(EX, W) is range from 79.8 to 85.0 dB(A). L_(EX, W) among all posts were greater than 80 dB(A) except naphtha processing operators. The result of pure tone hearing threshold test showed that the prevalence of high-frequency hearing loss among workers exposed to noise was 12.8%. According to the classification of noise operation according to the maximum value of L_(EX, W), the operator for styrene dry gas combined unit is the only post that is extremely dangerous in the department of chemical, the other posts in the department of chemical and all posts in the department of public works are exposed to light and medium noise hazards, and 62.5% the external operators of the oil refining department are under heavy and extremely hazardous. According to the evaluation result of ISO 1999:2013, the risk of high-frequency hearing loss among workers in crude distillation unit, hydrogen production unit and the electricians of electrical system is high. The measured median of noise-induced pernament threshold shift(NIPTS) among male workers in different workstation was higher than the predicted median of NIPTS of ISO 1999, and the difference was statistically significant(P<0.01), and the predicted values for four-fifths of positions were more than 10 dB lower than the measured value. CONCLUSION: The risk of noise induced hearing loss of workers in petrochemical enterprises is high.

RESUMO

BACKGROUND: Chinese citizens traveling abroad bring back imported malaria cases to China. Current malaria diagnostic tests, including microscopy and antigen-detecting rapid tests, cannot reliably detect low-density infections. To complement existing diagnostic methods, we aimed to develop a new loop-mediated isothermal amplification (LAMP) assay to detect and identify Plasmodium falciparum in Chinese travelers returning from Africa. METHODS: We developed a miniaturized LAMP assay to amplify the actin I gene of P. falciparum. Each reaction consumed only 25% of the reagents used in a conventional LAMP assay and the same amount of DNA templates used in nested PCR. We evaluated this LAMP assay's performance and compared it to microscopy and a nested PCR assay using 466 suspected malaria cases imported from Africa. We assessed the sensitivity of the new LAMP assay using cultured P. falciparum, clinical samples, and a plasmid construct, allowing unprecedented precision when quantifying the limit of detection. RESULTS: The new LAMP assay was highly sensitive and detected two more malaria cases than nested PCR. Compared to nested PCR, the sensitivity and specificity of the novel LAMP assay were 100% [95% confidence interval (CI) 98.5-100%] and 99.1% (95% CI 96.7-99.9%), respectively. When evaluated using serial dilutions of the plasmid construct, the detection limit of the new LAMP was as low as 102 copies/µL, 10-fold lower than PCR. The LAMP assay detected 0.01 parasites/µL of blood (equal to 0.04 parasites/µL of DNA) using cultured P. falciparum and 1-7 parasites/µL of blood (4-28 parasites/µL of DNA) in clinical samples, which is as good as or better than previously reported and commercially licensed assays. CONCLUSION: The novel LAMP assay based on the P. falciparum actin I gene was specific, sensitive, and cost-effective, as it consumes 1/4 of the reagents in a typical LAMP reaction.

RESUMO

This study aims to establish the model of the cryptocurrency price trend based on a financial theory using the Long Short-Term Memory (LSTM) networks model with multiple combinations between the window length and the predicting horizons. The Random Walk model is also applied with different parameter settings. The object of this study is the cryptocurrency and medical issues, primarily the Bitcoin and Ethereum and the COVID-19. Quantitative analysis is adopted as the method of this dissertation. The research tool is Python programming language, and the TensorFlow package is employed to model and analyze research topics. The results of this study show the limitations of the LSTM and Random Walk model for price prediction while demonstrating the different characteristics of both models with different parameter settings, providing a balance between the model's accuracy and the model's practicality.

RESUMO

To form a unified configuration and information management platform, FCCMS (financial center configuration management system) will integrate and sort information based on various configuration data and relationships as well as integrate processes and permissions. However, the most serious issue that data centers are currently facing is how to effectively manage these infrastructures. For various infrastructures, the data center currently uses a decentralized operation and maintenance management model. When an infrastructure fails due to inexperienced configuration management, this mode is not conducive to quickly locating and resolving the problem. A detection method of RFCO (random forest algorithm based on clustering optimization) is proposed, and an appropriate tree is selected from RF to integrate, so as to achieve the best effect. In this paper, the target matching algorithm based on FSL (few-shot learning) is deeply studied, and the target detection model is applied to the target matching and positioning task by using the ML method. The performance of the algorithm is tested by experiments on relevant datasets to verify the effectiveness of the algorithm in various scenarios.

RESUMO

Photoplethysmography (PPG) is an important signal which contains much physiological information like heart rate and cardiovascular health etc. However, PPG signals are easily corrupted by motion artifacts and body movements during their recordings, which may lead to poor quality. In order to accurately extract cardiovascular information, it is necessary to ensure high PPG quality in these applications. Although there are several existed methods to get the PPG signal quality, those algorithms are complex and the accuracies are not very high. Thus, this work proposes a deep learning network for the signal quality assessment using the STFT time-frequency spectra. A total of 5804 10s signals are preprocessed and transformed into 2D STFT spectra with 250 × 334 pixels. The STFT figures are as the input of the CNN networks, and the model gives the result as good or bad quality. The model accuracy is 98.3% with 98.9% sensitivity, 96.7% specificity, and 98.8% F1-score. And the heart rate error is much reduced after classification with the reference of ECG signals. Thus, the proposed deep learning approaches can be useful in the classification of good and bad PPG signals. As far as we know, this is the first article using deep learning methods combined with STFT time-frequency spectra to get the signal quality assessment of PPG signals.

RESUMO

BACKGROUND: Family caregivers need to be supported in caring for patients at the end of life, but practical tools to assess their support needs have been missing in China. So this study aimed to culturally adapt and validate the Carer Support Needs Assessment Tool (CSNAT). METHODS: Cross-cultural adaptation of the original CSNAT for a Chinese setting was performed according to Brislin's translation guidelines. A pilot study was conducted with 15 Chinese family caregivers of cancer patients receiving hospice home care and 5 medical staff. A cross-sectional survey of 205 family caregivers was conducted from December 2018 to May 2019 at a home-based hospice care institute in Shenzhen, China. The validation procedure comprised the establishment of (1) content validity by a group of six experts; (2) face validity by 15 family caregivers; (3) criterion validity by calculating Spearman's correlations between the CSNAT and caregiving burden, caregiving preparedness and quality of life scales; (4) internal consistency using Cronbach's alpha. RESULTS: The CSNAT demonstrated good face validity and good content validity. CSNAT scores showed clear positive correlations with caregiving burden and negative correlations with preparedness for caregiving and quality of life. Internal consistency was high (Cronbach's alpha = 0.899), although such reliability testing is not recommended for this tool. CONCLUSIONS: The Chinese version of the CSNAT is a valid tool that is appropriate for identifying needs of family caregivers of cancer patients in home-based hospice care.

RESUMO

To promote the rational application of nitrogen fertilizer for winter wheat under rice-wheat rotation in the Yangtze River Basin, we examined the effects of nitrogen application rates (0, 120, 210, 300 kg·hm-2, expressed as N0, N1, N2, and N3 respectively) on soil nitrate content, nitrogen balance of soil-plant system and yield. The results showed that soil nitrate content increased with increasing nitrogen application rates. Under different nitrogen application treatments, all the nitrate was significantly transfered to the 60 cm soil layer till jointing stage. After jointing stage, topdressing nitrogen significantly increased nitrate content in 0-40 cm soil layer under N1 and N2 treatments and that in the 0-60 cm soil layer under N3 treatment. Soil nitrate mainly accumulated in the 0-40 cm soil layer in the mature stage. Results from nitrogen balance analysis showed that nitrogen absorption, residue and loss varied across different growth stages of wheat, with the period from overwintering to jointing being the principal time of apparent nitrogen loss. The amount of plant nitrogen accumulation, inorganic nitrogen residue and soil nitrogen apparent loss all positively correlated with the nitrogen application rate. Based on the comprehensive analysis through Coase principle and marginal revenue of environmental economics, the optimum nitrogen application rate for production, ecology and economic benefits of winter wheat under rice-wheat rotation was 250 kg·hm-2, and the ratio of base fertilizer to jointing fertilizer was 5:5, while the corresponding grain yield was 6840 kg·hm-2.

RESUMO

In pedestrian inertial navigation, multi-sensor fusion is often used to obtain accurate heading estimates. As a widely distributed signal source, the geomagnetic field is convenient to provide sufficiently accurate heading angles. Unfortunately, there is a broad presence of artificial magnetic perturbations in indoor environments, leading to difficulties in geomagnetic correction. In this paper, by analyzing the spatial distribution model of the magnetic interference field on the geomagnetic field, two quantitative features have been found to be crucial in distinguishing normal magnetic data from anomalies. By leveraging these two features and the classification and regression tree (CART) algorithm, we trained a decision tree that is capable of extracting magnetic data from distorted measurements. Furthermore, this well-trained decision tree can be used as a reject gate in a Kalman filter. By combining the decision tree and Kalman filter, a high-precision indoor pedestrian navigation system based on a magnetically assisted inertial system is proposed. This system is then validated in a real indoor environment, and the results show that our system delivers state-of-the-art positioning performance. Compared to other baseline algorithms, an improvement of over 70% in the positioning accuracy is achieved.

RESUMO

In this paper, we present a novel thermoresistive gas flow sensor with a high-yield and low-cost volume production by using front-side microfabricated technology. To best improve the thermal resistance, a micro-air-trench between the heater and the thermistors was opened to minimize the heat loss from the heater to the silicon substrate. Two types of gas flow sensors were designed with the optimal thermal-insulation configuration and fabricated by a single-wafer-based single-side process in (111) wafers, where the type A sensor has two thermistors while the type B sensor has four. Chip dimensions of both sensors are as small as 0.7 mm × 0.7 mm and the sensors achieve a short response time of 1.5 ms. Furthermore, without using any amplification, the normalized sensitivity of type A and type B sensors is 1.9 mV/(SLM)/mW and 3.9 mV/(SLM)/mW for nitrogen gas flow and the minimum detectable flow rate is estimated at about 0.53 and 0.26 standard cubic centimeter per minute (sccm), respectively.

RESUMO

OBJECTIVES: Limited therapeutic options exist for treating severe infections caused by multidrug-resistant (MDR) and extensively drug-resistant Gram-negative bacteria (GNB). In this study, the activity of colistin (COL) as monotherapy and in combination with other antibiotics against Acinetobacter baumannii in vitro was investigated. In addition, the efficacy of intravenous colistimethate sodium (CMS) was evaluated in a murine model of urinary tract infection (UTI) induced by MDR Escherichia coli. METHODS: Minimum inhibitory concentration (MIC), Monte Carlo simulation, fractional inhibitory concentration index (FICI), time-kill study and erythrocyte lysis assay were applied to evaluate the effect and cytotoxicity of COL, meropenem, imipenem, doripenem (DOR) and sulbactam alone and in combination. For the in vivo experiment, determination of the bacterial burden and histopathological examination were performed to evaluate the efficacy of CMS against UTI. RESULTS: Of 106 A. baumannii isolates, 104 (98.1%) were susceptible to COL. In the chequerboard assay, COL + DOR showed the highest rate of synergism (60%). No antagonism or cytotoxicity was observed. All COL-based combinations were able to inhibit or slow bacterial re-growth in a time-kill assay. In an in vivo activity study, intravenous CMS reduced not only the bacterial load but also inflammation and maintained structural integrity of infected bladders and kidneys. CONCLUSION: The effectiveness of COL alone in vitro and in vivo suggested that intravenous CMS will be an effective and available therapeutic strategy for UTI due to MDR-GNB. In-depth in vitro tests demonstrated that COL + DOR could be an attractive option, especially when the COL MIC is ≥1 µg/mL.

RESUMO

Culture-dependent and -independent methods were employed to determine the impact of carboxyl-functionalized single-walled carbon nanotubes (SWNTs) on fungal and bacterial soil microbial communities. Soil samples were exposed to 0 (control), 250, and 500 µg of SWNTs per gram of soil. Aliquots of soil were sampled for up to 14 days for culture-dependent analyses, namely, plate count agar and bacterial community level physiological profiles, and culture-independent analyses, namely, quantitative real-time polymerase chain reaction (qPCR), mutliplex-terminal restriction fragment length polymorphism (M-TRFLP), and clone libraries. Results from culture-independent and -dependent methods show that the bacterial soil community is transiently affected by the presence of SWNTs. The major impact of SWNTs on bacterial community was observed after 3 days of exposure, but the bacterial community completely recovered after 14 days. However, no recovery of the fungal community was observed for the duration of the experiment. Physiological and DNA microbial community analyses suggest that fungi and bacteria involved in carbon and phosphorus biogeochemical cycles can be adversely affected by the presence of SWNTs. This study suggests that high concentrations of SWNTs can have widely varying effects on microbial communities and biogeochemical cycling of nutrients in soils.

RESUMO

The improvement of DNA's bioactivities by altering their structure is meaningful for their biological applications, ranging from DNA condensation study to gene therapeutic research. In this study, we treated the plasmid DNA with alkali and investigated the structure and the condensation efficiency of the alkali-treated DNA. We noticed that the alkali treatment could significantly increase the DNA condensation efficiency with spermidine and polyethylenimine (PEI). In addition, due to the improved interactions between the alkali-treated DNA and PEI, gene transfection experiments could be performed in the presence of less PEI. This research can contribute to the creation of a universal method to enhance the interaction between DNA and gene delivery vectors by alkali treatment, and should have significant potential in the field of gene therapy.

RESUMO

Chemicals in the environment often exist as mixtures and at low doses, whose ecotoxicological diagnosis and risk assessment are hard to be done with the help of the research results on the ecotoxicity of high-level exposure of single chemicals. This paper reviewed the research advances in ecotoxicology and risk assessment of low-dose chemical mixture contamination, with the focus on the molecular toxicological methods and the risk assessment approaches and schemes of simple and complex mixtures. Comments were given on the development directions of low-dose chemical mixture research, including the necessity of searching for sensitive endpoints, involving the approaches of other disciplines, increasing data accumulation, and setting up a comprehensive and identical risk assessment system.
